SUMMARY:Bioengineering Seminar Series - Dr. Adele Moatti
LOCATION:Benedum Hall 157\, 3700 O'Hara Street\, Pittsburgh\, PA 15261
DESCRIPTION:Dr. Adele Moatti\, Assistant Professor in the Department of Ot
olaryngology–Head and Neck Surgery at the University of Pittsburgh School
of Medicine and Adjunct Assistant Professor of Biomedical Engineering at
Carnegie Mellon University\, will present "Rebuilding the Translational
Pipeline for Inner Ear Therapies."\n\nAbstract\n\nA major barrier in inne
 r ear therapeutics is not a single limitation\, but a broken pipeline\, w
here clinically relevant models\, targeting strategies\, and delivery mec
hanisms fail to align. Here\, we rebuild this pipeline by integrating com
plementary translational advances. We establish a porcine model of cochle
ar synaptopathy that captures key features of human pathology\, enabling
meaningful evaluation of therapeutic interventions. In parallel\, we gene
rate a comprehensive map of AAV expression in the pig inner ear\, providi
ng a critical bridge between mouse studies and clinically relevant gene t
herapy design. We further demonstrate that extracellular vesicles (EVs) c
an be harnessed to enhance safe local AAV delivery\, improving transducti
on efficiency while reducing immune responses. Finally\, we uncover an un
expected biological lever of drug delivery: sensory-driven formulation ef
fects. We show that modulation of taste receptor signaling can extend dru
g retention within the inner ear\, revealing a new axis for therapeutic c
ontrol. Together\, these advances shift the paradigm from isolated soluti
ons to an integrated\, translatable framework for inner ear gene and d\n\
