 Parkinson’s Disease: Lipid Droplets\, Neuroinflammation\, and\nSynaptic R
 emodeling\n\nBy\nSaid S. Salehe\n\nBrain aging and neurodegenerative diso
rders such as Parkinson's disease (PD) share several hallmarks\,\nincludi
ng chronic neuroinflammation\, pathological lipid-droplet (LD) accumulati
on\, and synaptic alterations.\nHowever\, the upstream regulators of thes
e processes remain poorly defined. This dissertation identifies noncanoni
cal\nroles for dynamin-related protein 1 (DRP1)\, best known for mediatin
 g mitochondrial fission\, in\nregulating lipid metabolism\, neuroinflamma
tion\, and synaptic integrity.\nAcross models of aging\, lipopolysacchari
 de (LPS)-induced inflammation\, and α-synuclein\noverexpression\, LD accu
mulation was greater in microglia than in other cell types. Microglial DR
 P1 abundance\ncorrelated positively with LD burden\, whereas partial DRP1
knockout or pharmacological inhibition reduced\nLD accumulation and infl
ammatory gene expression. Mechanistically\, DRP1 deficiency suppressed LP
 Sinduced\nlong-chain acyl-CoA synthetase 1 (Acsl1) expression via NF-κB s
ignaling. Inhibiting NF-κB or ACSL\nactivity likewise decreased LD accumu
lation in primary microglia. In human postmortem substantia nigra\,\nACSL
1 expression and PLIN2-positive LD burden were elevated relative to non-P
 D tissue\, with increa\n\nhttp://next-to-u.app/events/893f3f45-ee50-42a1-
