SUMMARY:BME7900 Seminar: Dr. Wilbur A. Lam (Emory)
LOCATION:Weill Hall 226
DESCRIPTION:Engineering Microsystems to Investigate the Mechanobiology of
Blood in Health and Disease\n\nBlood is fundamentally mechanical in natur
e\, comprising diverse cellular subtypes navigating complex hydrodynamic
environments. Perturbations in these biomechanical dynamics directly driv
e hematologic and cardiovascular pathologies. This presentation highlight
 s our lab's multidisciplinary approach—combining microfluidics\, biophysi
cs\, and hematology—to bridge microscale mechanobiology with clinical dia
gnostic and therapeutic translation.\n\nFirst\, we explore how bioenginee
red microfluidic systems model pathological mechanobiolohy. In sickle cel
l disease\, we describe how reduced erythrocyte deformability in and of i
tself causes aberrant cell margination toward microvascular walls. Comput
ational simulations and endothelialized microfluidic systems pioneered by
our laboratory reveal that marginated\, stiffened red blood cells induce
localized wall shear stress fluctuations\, triggering pro-inflammatory e
ndothelial gene expression via mechanotransduction—a mechanism distinct f
rom the canonical microvascular obstruction that oversimplifies sickle ce
ll pathophysiology.\n\nNext\, using advanced hydrogel-based microvasculat
 ure-on-a-chip platforms\, we investigate long-term vascular dysfunction\,
endothelial barrier permeability\, and thrombo-inflammation over weeks t
o months\, demonstrating how targeted therapeutics like tPA facilitate cl
ot resolution to restore endothelial function after thrombosis. Our l\n\n
